Use a garage or paid car park outside the old town, in or near the modern Murat district — the orderly 19th-century grid of shopping streets below Bari Vecchia. From there both the old town and the seafront are an easy walk. Avoid leaving valuables visible in a parked car, a sensible precaution in any busy port city.
Bari is one of the main Adriatic ferry ports, with services across to Greece (including Igoumenitsa and Patras), Albania (Durrës), Croatia and Montenegro. If you're catching a ferry, the port is just north of the centre, and you can drive a car onto many of the routes. Check in well ahead, as vehicle ferries have strict boarding times and you'll want to factor in the ZTL and parking if exploring the city first.
Yes — it's the regional capital and transport hub, well placed for the rest of Puglia. The trulli houses of Alberobello, the white town of Ostuni, the baroque city of Lecce and the beaches of the Adriatic coast are all reachable by car or regional train. A car is useful for the smaller towns and the coast, parked outside Bari's ZTL when you're in the city itself.
Not for the city — the old town and the grid-planned Murat district are walkable. A car helps for exploring Puglia — Alberobello, Ostuni, Lecce, the Adriatic beaches — driven out from a peripheral car park. If you're taking a ferry east, you can bring the car aboard many routes from the port.
